An antibody specific for PSMD10 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPSMD10 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for PSMD10 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PSMD10 bound in the initial step. The color development is stopped and the intensity of the color is measured. Product Overview:The 26S proteasome is a mµLticatalytic proteinase complex with a highly ordered structure composed of 2 complexes, a 20S core and a 19S regµLator. Proteasomes are distributed throµghout eukaryotic cells at a high concentration and cleave peptides in an ATP/ubiquitin-dependent process in a non-lysosomal pathway. An essential function of a modified proteasome, the immunoproteasome, is the processing of class I MHC peptides. PSMD10 encodes a non-ATPase subunit of the 19S regµLator. Two transcripts encoding different isoforms have been described. Pseudogenes have been identified on chromosomes 3 and 20.The predicted 226-amino acid p28 protein contains 5 ankyrin repeats, which are thoµght to function in protein-protein interactions.
