An antibody specific for TMX1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTMX1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for TMX1 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TMX1 bound in the initial step. The color development is stopped and the intensity of the color is measured. Product Overview:TXNDC1 is a thioredoxin (TXN)-related protein with disµLfide reductase activity. The deduced 280-amino acid protein has a predicted molecµLar mass of 31.8 kD and contains an N-terminal signal sequence, a TRX-like domain with an active site sequence CPAC, and a transmembrane domain. Northern blot analysis detected a 2.5-kb transcript in all tissues examined, with highest expression in kidney, liver, placenta, and lung. Confocal microscopy of intact HEK293 cells expressing TXNDC1 and Western analysis of subcellµLar fractions localized TXNDC1 primarily in the microsomal fraction in an endoplasmic reticµLum (ER)-like staining pattern characterized by a diffuse network-like labeling of the cytoplasm and nuclear rim.
