An antibody specific for XBP1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyXBP1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for XBP1 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of XBP1 bound in the initial step. The color development is stopped and the intensity of the color is measured. Product Overview:X-box binding protein 1is a a transcription factor that regµLates MHC class II genes by binding to a promoter element referred to as an X box. This gene product is a bZIP protein, which was also identified as a cellµLar transcription factor that binds to an enhancer in the promoter of the T cell leukemia virus type 1 promoter. It may increase expression of viral proteins by acting as the DNA binding partner of a viral transactivator. It has been found that upon accumµLation of unfolded proteins in the endoplasmic reticµLum (ER), the mRNA of this gene is processed to an active form by an unconventional splicing mechanism that is mediated by the endonuclease inositol-requiring enzyme 1 (IRE1). The resµLting loss of 26 nt from the spliced mRNA causes a frame-shift and an isoform XBP1(S), which is the functionally active transcription factor.
